NPM1 Antibody (NA24) Summary
BALB/C mice were immunized with GST fusion protein containing the N-terminal part of nucleophosmin fused to 14 amino acid of ALK protein.
This antibody with a protein of 427 kD. This antibody is highly specific to dystrophin and shows no cross-reaction with C-protein, ?-actin and human muscle spectrin. Dystrophin is a member of spectrin/alpha actinin family of actin-binding protein.

Background
Nucleophosmin (NPM) is a ubiquitously expressed oestrogen-regulated nucleolar phosphoprotein frequently overexpressed in a variety of human malignancies. It is a multifunctional protein, modulating diverse molecular functions which include ribosome biogenesis and centrosome duplication etc. It also functions as a molecular chaperone that prevents proteins from aggregating in the crowded environment of the nucleolus. It also modulates, both positively and negatively, the activity of some tumour suppressor genes such as p53 and Arf, and it is upregulated in response to stress stimuli including DNA damageand hypoxia. It continuously shuttles between the nucleus and cytoplasm. The NPM gene which is mapped to human chromosome 5q35 is involved in several tumour-associated chromosome translocations, which have resulted in the formation of fusion proteins that retain the amino terminus of NPM, including NPM-ALK4, NPM-RAR5 and NPM-MLF1. The protein is directly involved in cancer pathogenesis, as the NPM1gene is mutated in a number of human haematopoietic malignancies, such as promyelocytic leukaemia, anaplastic large cell lymphoma, acute myeloid leukaemia, chronic myelogenous leukaemia and MDS. However its role in oncogenesis is controversial as NPM has been attributed with both oncogenic and tumour suppressive functions.
