NB100-63087 detects mouse myeloid cell leukaemia-1 (Mcl-1), an anti-apoptotic member of the Bcl-2 family of proteins. It has a pro-survival role in the early stages of apoptosis, and is thought to prevent cytochrome c release from mitochondria. Mcl-1 is also essential for embryogenesis and for the development and maintenance of B and T lymphocytes. Mcl-1 is a highly regulated protein induced by a wide range of survival signals, including cytokines and growth factors. During apoptosis, Mcl-1 is rapidly degraded by the proteasome pathway, and is also readily cleaved by caspases to produce a potent cell death promoting protein. Two transcript variants encoding distinct isoforms of this protein have been identified, a longer gene product (isoform 1) which enhances cell survival by inhibiting apoptosis, and a shorter gene product (isoform 2) which promotes apoptosis and is death-inducing.
