    Specificity
    The specificity of this antibody is equivalent to that of CD41 monoclonal antibodies, and it detects the Glycoprotein IIb-IIIa complex on the surface of platelets and megakaryocytes. It is not reactive with T and B cells or with other haemopoietic cells. The antibody precipitates a complex of molecular weights 122kD and 112kD when reduced and 87kD and 140kD when not reduced1. The IIb-IIIa complex is a member of the integrin superfamily of adhesive receptors and mediates platelet-platelet aggregation. This complex recognizes the Arg-Gly-Asp (RGD) tripeptide sequence in adhesive glycoproteins such as fibronectin, fibrinogen and von Willebrand Factor thereby facilitating linkage between adjacent platelets.

    Application Notes
    This antibody is useful in biochemical studies of the Glycoprotein IIb-IIIa complex and the enumeration of platelets. The antibody specifically inhibits ADP and collagen induced aggregation and release of ATP, and ADP induced platelet fibrinogen binding1. The antibody may also be used for immunoperoxidase staining of formalin fixed cell suspensions and frozen tissue sections. Flow cytometry and indirect immunofluorescence. Dilute with isotonic buffer. Use 50 ul of diluted antibody per 1x10E6 platelets in 100 ul of buffer. 2. Indirect immunoperoxidase staining – the final dilution will depend on the assay conditions and detection system employed. However, a dilution of at least 1:25 will be applicable to most commonly used systems.
